Biodegradable polymeric nanofibers for tissue engineering and drug delivery applications


Tissue engineering is the remedy for regeneration of damaged tissue by providing biodegradable polymers with cells and growth factor. The preparation of Biomaterial which can mimic similar to physiological 3 dimensional tissues is still challenging. Biodegradable Polymers are used for development of tissue engineering constructs in the form of sponges, films and macro porous scaffolds which not influencing  the cell fate process such as cell differentiation, migration and proliferation. The extra cellular matrix is a component of all mammalian tissues and consists of a network of fibrous proteins, elastin and collagens, embedded in a viscoelastic gel rich in high molecular weight anionic polymers known as proteoglycans. Polymer nanofiber was fabricated by electro spinning of a polymer solution have been drawing great attention for the biomedical applications. The electrospun materials have the nanofibrous morphology which can mimic almost the natural extracellular matrix (ECM), which is composed of three-dimensional networks of nanosized fibrous proteins such as collagen. Biodegradable Polymers fabricates in form of fibers in nanosize that cause more favorable microenvironment for cells.


We prepared the PLGA/PPG nanofibers by elctrospinning technique.PLGA (85:15)/PLGA (75.25) nanofibers have hydrophobic and it can be minimized by addition of PPG to give better hydrophilitcity for cell adhesion for tissue engineering construct. The morphology of the electros pun fibers of composite of PLGA and PPG was observed using scanning electron Microscopy (SEM).The contact angle of these nanofibers scaffold and protein adsorption and release studies on that scaffold have done.

The results proved that the small amount of Polypropylene Glycol polymer to the Poly Lactic Glycolic acid (PLGA 85:15) improves drastically the hydrophilic of elctrospun nanofibers. The addition of small amount hydrophilic polymer to the biodegradable hydrophobic polymer increase the hydrophilic property and used for nanofiber based tissue engineering constructs and drug delivery applications.
